Phenobarbitone acts on GABAA receptors, increasing synaptic inhibition. This has the effect of elevating seizure threshold and reducing the spread of seizure activity from a seizure focus. Phenobarbitone may also inhibit calcium channels, resulting in a decrease in excitatory transmitter release.

Coumarin anticoagulants , alcohol and other CNS depressants , oral contraceptives , griseofulvin , rifampicin , phenytoin , metronidazole , doxycycline , chloramphenicol , telithromycin , antidepressants , St.John's wort , antipsychotics , nucleoside reverse transcriptase inhibitors , protease inhibitors , eplerenone , voriconazole , other anticonvulsants , digoxin , folic acid , methylphenidate .

A small number of people being treated with anti-epileptics such as phenobarbitone have had thoughts of harming or killing themselves. If at any time you have these thoughts, immediately contact your doctor.
Potentially life-threatening skin rashes (Stevens-Johnson syndrome, toxic epidermal necrolysis) have been reported with the use of Phenobarbitone, appearing initially as reddish target like spots or circular patches often with central blisters on the trunk.
These potentially life-threatening skin rashes are often accompanied by flu-like symptoms. The rash may progress to widespread blistering or peeling of the skin.
The highest occurence of serious skin reactions is within the first week of treatment. If you have developed Stevens-Johnson syndrome or toxic epidermal necrolysis with the use of Phenobarbitone, you must not be restarted on Phenobarbitone at any time.
