Benign Prostatic Hyperplasia
Dutasteride inhibits the conversion of testosterone to 5a-dihydrotestosterone (DHT). DHT is the androgen primarily responsible for the initial development and subsequent enlargement of the prostate gland. Testosterone is converted to DHT by the enzyme 5a-reductase, which exists as 2 isoforms, type 1 and type 2. The type 2 isoenzyme is primarily active in the reproductive tissues, while the type 1 isoenzyme is also responsible for testosterone conversion in the skin and liver. Dutasteride is a competitive and specific inhibitor of both type 1 and type 2 5a-reductase isoenzymes, with which it forms a stable enzyme complex. Dissociation from this complex has been evaluated under in vitro and in vivo conditions and is extremely slow. Dutasteride does not bind to the human androgen receptor.

It is used to treat men with an enlarged prostate (benign prostatic hyperplasia) ; A non-cancerous growth of the prostate gland , caused by producing too much of a hormone called dihydrotestosterone.
Do not take • If you are allergic to dutasteride, other 5-alpha reductase inhibitors, soya, peanut or to any of the other ingredients of this medicine • If you have a severe liver disease.
Lower urinary tract symptoms of BPH can be indicative of other urological diseases, including prostate cancer. Patients should be assessed to rule out other urological diseases prior to treatment with Dutasteride. Patients with a large residual urinary volume and/or severely diminished urinary flow may not be good candidates for 5a-reductase inhibitor therapy and should be carefully monitored for obstructive uropathy.
Dutasteride is absorbed through the skin, therefore, women, children and adolescents must avoid contact with leaking capsules . If contact is made with leaking capsules, the contact area should be washed immediately with soap and water.
Men treated with dutasteride should not donate blood until at least 6 months have passed following their last dose to prevent pregnant women from receiving dutasteride through blood transfusion .
Caution is advised when alpha adrenergic blocking agents including tamsulosin are coadministered with PDE5 inhibitors. Alpha adrenergic blockers and PDE5 inhibitors are both vasodilators that can lower blood pressure. Concomitant use of these two drug classes can potentially cause symptomatic hypotension .
